50X Denhardt's Solution Recipe

  • 1 g Ficoll 400
  • 1 g polyvinylpyrrolidone (PVP)
  • 1 g bovine serum albumin (BSA, Fraction V)
  • Ultrapure or deionized water to a final volume of 100 mL

Denhardt's solution is commonly added to prehybridization and hybridization buffers used in Northern and Southern blotting. Ficoll 400, polyvinylpyrrolidone, and bovine serum albumin help occupy nonspecific binding sites on the membrane, reducing background caused by probes or detection reagents binding outside the intended nucleic acid target.

This conventional 50X stock contains 1% (w/v) each of Ficoll 400, PVP, and BSA. Diluting it to 1X gives a final concentration of 0.02% (w/v) for each component.

Use this stock as a convenient supplement when preparing prehybridization or hybridization buffers, diluting it to the final Denhardt's concentration specified by the protocol.

  1. Add approximately 80 mL of ultrapure or deionized water to a suitable vessel.
  2. Add 1 g of Ficoll 400 and stir until dissolved.
  3. Add 1 g of polyvinylpyrrolidone (PVP) and continue stirring until dissolved.
  4. Add 1 g of bovine serum albumin (BSA, Fraction V) and mix gently until the solution is uniform, minimizing foaming.
  5. Add ultrapure or deionized water until the final total volume is 100 mL.
  6. Mix the solution thoroughly.
  7. Sterilize by filtration through a 0.22 micrometer membrane if required by the intended application.
  8. Divide into suitable aliquots and store according to the requirements of the protocol.

Denhardt's solution is a blocking supplement rather than a complete hybridization buffer. Add it to a hybridization buffer formulated with the salts, detergents, buffering agents, nucleic acid blockers, and other components required by the chosen protocol.
